Foodborne diseases, caused by pathogenic bacteria, have become an important social issue in the field of food safety. It presents a widespread and growing threat to human health in both developed and developing countries. As such, techniques for the detection of foodborne pathogens and waterborne pathogens are urgently needed to prevent the occurrence of human foodborne infections. Although traditional culture-based bacterial isolation and identification are the "gold standard" methods with high preciseness, their drawbacks in time-consuming are inadequate for rapid detection of pathogen to reduce foodborne disease occurrence. Fortunately, with the development of biotechnologies and nanotechnologies, various kinds of new technologies for rapid detection of pathogens have been developed so far, such as nucleic acid-based methods, antibody-based methods, and aptamer-based assays. In this chapter, we summarized the principles and the application of some recent rapid detection technologies for pathogenic bacteria. Moreover, the advantages and disadvantages of the established and emerging rapid detection methods are addressed here.

Для молекулярной диагностики инфекций картофеля Y-вирусом и вирусом скручива-ния листьев картофеля (ВСЛК) разработаны иммунохроматографические тест-системы. Чтобы размножить антиген -капсидный белок труднодоступного флоэмно-ограничен-ного ВСЛК, был создан бинарный вектор, содержащий кДНК рекомбинантной РНК то-бамовируса, в которой ген капсидного белка был заменен на ген белка оболочки ВСЛК. Рекомбинантная тобамовирусная РНК упаковывалась белком оболочки ВСЛК в сфери-ческие вирусные частицы. Химерный вирус был инфекционен, а выход его и белка обо-лочки ВСЛК при выделении был в 800 раз выше, чем у дикого типа. К химерному вирусу были получены антитела для лабораторного и полевого анализа инфекции картофеля. На основании опыта разработки и применения диагностических тест-систем предлага-ется тактика рациональной массовой лабораторной и практической диагностики вирус-ных заболеваний организмов на молекулярном уровне. Ключевые слова: Y-вирус картофеля, вирус скручивания листьев картофеля, химерный вирус, иммунохроматография, инфекция, молекулярная диагностика.Молекулярная диагностика инфекций является частным случаем поиска целевой нуклеотидной по-следовательности и/или целевого антигена. В об-щем случае молекулярная диагностика не зависит от биологической природы патогена и организма хозяина, меняется только метод подготовки об-разца к анализу [1].Современная массовая диагностика основана на двух принципах молекулярного взаимодействия: на молекулярной гибридизации комплементарных гетероциклических оснований нуклеиновых кислот и на узнавании антигена антителом. В недалеком будущем геномное секвенирование и генетические паспорта организмов станут обычными. В этом слу-чае молекулярная диагностика сможет не только устанавливать окончательный диагноз, но и, воз-можно, позволит узнать, как возникло заболевание.В лабораторной диагностике фитопатогенов наиболее часто используются иммуноферментные и иммунохимические анализы (ИФА, ИХА), им-мунохроматографические тест-системы (ИХТС), иммуночипы; ДНК(РНК)-зонды, ПЦР и ОТ-ПЦР (обратная транскрипция-полимеразная цепная ре-акция), ДНК(олигонуклеотиды)-чипы. Для дости-жения предельной чувствительности диагностического анализа, например, в технологии ОТ-ПЦР-МГА (молекулярный-гибридизационный анализ)-ИФА, системы усиления сигнала комбинируют [2].Доминирующими в практической лабораторной диагностике вирусов растений являются технологии иммуноферментного анализа, молекулярного гиб-ридизационного анализа и различные варианты полимеразной цепной реакции ввиду их высокой чувствительности, специфичности и производи-тельности. Для одновременного выявления не-скольких патогенов в одном образце предлагаются различные технические решения, из которых наи-более перспективным представляется развитие чиповой технологии.В МГУ на кафедре вирусологии биологиче-ского факультета и в НИИ ФХБ имени А.Н. Analytical parameters of immunochromatographic test-systems, such as the detection limit and working range, depend on many factors. However, the process of choosing the concentration and composition of reagents applied to test strip membranes, and choosing the membranes themselves has not been systematically described for commercially produced tests or for scientific studies. Here, we present the results of a study to determine how the properties of the test system components, using mycotoxin aflatoxin B1 as an example, affect its analytical performance. A comparison of different membranes used for immunochromatography was performed as well as reagent concentrations and composition of intermolecular conjugates. Optimization of the immunochromatographic system indicated a limit of aflatoxin B1 detection equal to 0.2 ng/mL. The effectiveness of the system in testing plant extracts with minimal (twice) dilution prior to analysis was determined and the final content of methanol in the course of immunoassay was 35%.
