Immunochemical characterisation of styrene maleic acid lipid particles prepared from Mycobacterium tuberculosis plasma membrane

Abstract: Membrane proteins of Mycobacterium tuberculosis (Mtb) can be targeted for the development of therapeutic and prophylactic interventions against tuberculosis. We have utilized the unique membrane-solubilising properties of the styrene maleic acid copolymer <styrene:maleic acid::2:1> (SMA) to prepare and characterise ‘styrene maleic acid lipid particles’ from the native membrane of Mtb (MtM-SMALPs). When resolved by SDS-PAGE and visualised with coomassie blue, the molecular weights of Mtb membrane (MtM) pr… Show more

“…It is relevant that the highly glycosylated M. tuberculosis proteins identified in the present work corresponded to Lipoproteins, a functionally several class of mycobacterial membrane proteins involved in cell invasion, colonization, evasion of host defense, cell biogenesis, adhesion, immunomodulation and transport over the membrane 7 , 9 , 31 . These include proteins like the lipoprotein LprA (Rv1270), LprH (Rv1418), GlnH (Rv0411c), LpqN (Rv05830, Mce lipoprotein Lprk (Rv0173), Mce1A (Rv0169), Mce2E (Rv0593), Mce4A (Rv3499c), Mce1B (Rv0170) and Mce1F (Rv0174) (Table 1 ).…”
