Transmissible venereal tumor (TVT) is a sexually transmitted, naturally occurring tumor of the canine family and often occurs in tropical and subtropical countries. The matrix metalloproteinases (MMPs) are endogenous proteases accountable for the degradation of extracellular matrix (ECM) components, such as collagen and other proteins including the basement membrane. MMPs play a vital role in the tumor metastasis and angiogenesis. Both MMP-2 and-7 strongly associated with the invasion and metastasis of different cancer types. This study aims to investigate the MMP-2 and-7 expression in naturally occurring TVT in 20 dogs using immunohistochemical methods. Immunohistochemically, we observed increased MMP-2 and-7 expressions in tumor cells. In addition, a positive correlation was determined between the tumor size and immunoexpressions of the markers indicating that both MMP-2 and-7 participate in the TVT pathogenesis. Matriks metalloproteinazlar (MMP)'ler endojen peptidazlar olup, ekstraselüler matriksin (ECM) kollagen ve bazal membran gibi diğer proteinlerini parçalama özelliğine sahiptirler. MMP'ler tümör metastaz ve angiogenezinde önemli rol oynarlar. MMP-2 ve MMP-7 değişik tip kanserlerde invazyon ve metastaz ile ilişkili bulunmuşlardır. Bu çalışmanın amacı, MMP-2 ve MMP-7'nin 20 adet köpekte doğal olarak şekillenmiş TVT olgusunda immunhistoimyasal olarak ekspresyonlarının incelenmesidir. İmmunohistokimyasal olarak TVT'yi oluşturan tümör hücrelerde MMP-2 ve MMP-7 aktivitesinde artış şekillendiği gözlendi. Ayrıca tümör büyüklüğü ile immunoekspresyonlar arasında pozitif korelasyon saptandı, bu sonuç, MMP-2 ve MMP-7'in TVT patojenezinde önemli bir rol oynadığını gösterdi.

In this study, we aimed to evaluate the proliferative, metastatic and apoptotic capacities of TVT cases, which are in various phases of development, by using immunohistochemical markers. The material of this study consisted of twelve female and six male dogs diagnosed with TVT brought to our department between 2007 and 2020 years. Diff-quick staining was applied to the smear taken from tumoral masses for cytological examinations. Tumoral tissues from dogs were fixed in a 10% neutral buffered formaldehyde solution. After routine tissue procedures Hematoxylin & Eosin stain was applied to the sections. Tissue sections were investigated under a light microscope and photographed. Immunohistochemical staining was performed on the tissues using the avidin-biotin immune peroxidase complex method. As a result of macroscopic, cytological and histopathological examinations, TVT positive cases were divided into three according to their developmental stages. While the expression of PCNA, MMP-9, mutant p53 and Bcl-2 increased significantly in progressive cases compared to regressive and stable cases, Bax expression increased significantly in regressive cases compared to progressive and stable cases. In conclusion, we thought that the mentioned markers are very useful for understanding the prognosis of TVT, the tumor aggressiveness and the survival of the malignant cells.

Monocarboxylate transporter 1 (MCT1) has been reported to be correlated wtih decreased survival and advanced stage of progression in a series of human tumor cells and primary cancers. Specifically, MCT1 has been documented to be involved in tumor progression, including invasion and migration. Here, we investigated the mechanism and effect of regulation of MCT1 on invasion and migration of nasopharyngeal carcinoma (NPC) cells. In the study, we firstly demonstrated that the expression of MCT1 in CNE2Z cells was obviously higher than that in HNE1 cells. Downregulation of MCT1 inhibited the invasion and migration in CNE2Z cells, upregulated the expression of E-cadherin, TIMP (tissue inhibitor of metalloproteinase)-2 and TIMP-1, and suppressed the expression of matrix metalloproteinases (MMP)-9 and MMP-2. Correspondingly, upregulation of MCT1 enhanced the invasive and migratory potential in HNE1 cells, increased the expression of MMP-9 and MMP-2, and downregulated the expression of E-cadherin, TIMP-2 and TIMP-1. The mechanistic study demonstrated that the effect of MCT1 might be correlated with PI3K/Akt signaling pathway. LY294002, a PI3K inhibitor, increased the inhibition of invasion and migration mediated by downregulation of MCT1 in CNE2Z cells. These findings collectively suggested that MCT1 might act as a new regulator to improve invasion and migration of NPC cells and be correlated with activating the PI3K/Akt pathway.
