Salmonella enterica subspecies enterica serovar abortus equi (S. Abortus equi) is the most common cause of abortion in mares. It has recently been found to cause abortion in donkeys more frequently in China. A novel virulent bacteriophage vB_SabS_Sds2 (hereafter designated as Sds2) was isolated from the feces of donkeys using a S. Abortus equi strain as a host. Phage Sds2 had an isometric polyhedral head and an uncontracted long tail, belonging to the Tequintavirus, Markadamsvirinae, Demerecviridae, Caudovirales. The genome of phage Sds2 was 114,770 bp, with a GC content of 40.26%. The genome contained 160 open reading frames (ORFs), and no ORFs were associated with pathogenicity, drug resistance, or lysogenization by sequence analysis. Both genome annotation and phylogenetic analysis indicated that phage Sds2 was highly similar to T5-like bacteriophages. Phage Sds2 could lyse 100% (30/30) of S. Abortus equi strains, 25.3% (24/95) of other serotypes of Salmonella strains, and 27.6% (8/29) of Escherichia coli strains using the double-layer agar plate method. The in vitro test showed that phage Sds2 had high bactericidal activity against S. Abortus equi at a wide range of MOIs. The in vivo test indicated that phage Sds2 had an inhibitory effect on abortion in mice challenged with S. Abortus equi. In general, phage Sds2 is a novel lytic phage with a wide host range and has the potential to prevent abortion caused by S. Abortus equi.

Inland aquaculture contributed by three major fish groups, including carps, tilapias, and catfish plays a vital role in global food security and nutrition, particularly in low and middle-income countries. However, the sustainable development of this sector is hampered by disease epidemics, especially those caused by bacteria such as Aeromonas species. At least eight pathogenic motile Aeromonas species (A. hydrophila, A. veronii, A. jandaei, A. caviae, A. sobria, A. bestiarum, A. dhakensis and A. schubertii) have been reported in aquaculture with some causing up to 100% mortality during disease outbreaks. Simultaneously, emerging multidrug-resistant Aeromonas due to a long-inappropriate use of antibiotics is alarming and highlights a global public health concern and negative socioeconomic impacts. Here, we provide a comprehensive overview of motile Aeromonas infections, antibiotic use and antimicrobial resistance of Aeromonas species. This contribution also highlights the non-antibiotic approaches (the solutions for preventing or treating of bacterial diseases without resorting to antibiotic use) to control motile Aeromonas infections. In addition to the current state of knowledge and limitations of each prophylaxis/therapy, perspectives for future research are discussed critically, including oral/immersion multivalent vaccines, microencapsulated synbiotics, exogenous metabolites, and novel lytic bacteriophage cocktails. Some emerging applicable nanotechnology themes such as nanovaccines, nanobioactive compounds, and nanobubbles are also included in this review. In

Salmonella enterica subspecies enterica serovar abortus equi (S. abortus equi) is a frequently reported pathogen causing abortion in mares. In this study, the preventive and therapeutic effects of phage PIZ SAE-01E2 against S. abortus equi in a mouse model of abortion was investigated. Phage PIZ SAE-01E2 was stable at different temperatures (4 ∼ 70 °C) and pH values (4 ∼ 10) and could lyse the majority of the tested Salmonella strains (25/28) of serotype O:4 and O:9. There is no lysogenic, toxin or antibiotic-resistant related gene in the genome of PIZ SAE-01E2. All of these characteristics indicated PIZ SAE-01E2 has potential for phage therapy. In in vivo experiments, 2 × 103 CFU/mouse of S. abortus equi ATCC 9842 was sufficient to lead to murine abortion (gestational day 14.5) within 48 h. A single dose of intraperitoneal inoculation of PIZ SAE-01E2 (108 PFU/mouse, MOI = 105) 1 h before or after S. abortus equi challenge provided effective protection to all pregnant mice (10/10). The bacterial loads in the placenta and uterus of the infected mice were both significantly decreased (< 102 CFU/g) after 24 h treatment with phage PIZ SAE-01E2 compared to that of control group (> 106 CFU/g). In addition, the levels of inflammatory cytokines in the placenta and blood of the mice in the phage administration groups were significantly reduced (P < 0.05) compared to that of the control group. Altogether, this indicated that PIZ SAE-01E2 showed potential to block abortions induced by S. abortus equi in vivo. Importance S. abortus equi is an important pathogen that can induce abortions in mares. Although S. abortus equi has been well controlled in Europe and United States due to strict breeding and health policies, it is still widespread in African and Asian countries and has proven difficult to control. In China, the abortions in donkeys caused by S. abortus equi have also been reported. So far, there is no commercial vaccine. Thus, exploiting alternative efficient and safety strategies to control S. abortus equi infection is essential. In this study, a new lytic phage PIZ SAE-01E2 infecting S. abortus equi was isolated and characteristics of PIZ SAE-01E2 indicated it has potential for phage therapy. A single dose of intraperitoneal inoculation of PIZ SAE-01E2 before or after S. abortus equi challenge provided effective protection to all pregnant mice. Thus, PIZ SAE-01E2 showed potential to block abortions induced by S. abortus equi in vivo.
