Immunization with different recombinant West Nile virus envelope proteins induces varying levels of serological cross-reactivity and protection from infection

Rebecca Weiß,
Leila Issmail,
Alexandra Rockstroh
et al.

Abstract: IntroductionWest Nile Virus (WNV) is a zoonotic flavivirus transmitted by mosquitoes. Especially in the elderly or in immunocompromised individuals an infection with WNV can lead to severe neurological symptoms. To date, no human vaccine against WNV is available. The Envelope (E) protein, located at the surface of flaviviruses, is involved in the invasion into host cells and is the major target for neutralizing antibodies and therefore central to vaccine development. Due to their close genetic and structural r… Show more

“…In this study, dengue IgMs were detected in 20% of negative RT-PCR samples suggesting arboviral positivity in them. Though a Zika IgM test might have been more appropriate in this case, cross-reactivity has been extensively reported in serological tests against these viruses making this method highly unspecific and difficult to be interpreted ( Musso and Desprès, 2020 ; da Rocha Queiroz Lima et al., 2021 ; Weiß et al., 2023 ; Zambrana et al., 2024 ). A proper dissection of arbovirus serology requires the gold standard for serological definition of specific positive sera, plaque reduction neutralization tests (PRNT).…”
