Immunity and immunopathology to viruses: what decides the outcome?

Abstract: PREFACE Many viruses infect humans and most are controlled satisfactorily by the immune system with limited damage to host tissues. Some viruses, however, do cause overt damage to the host, either in isolated cases or as a reaction that commonly occurs after infection. The outcome is influenced by properties of the infecting virus, the circumstances of infection and multiple factors controlled by the host. In this Review, we focus on host factors that influence the outcome of viral infection, including genetic… Show more

“…However, approximately 85% of patients respond with IL-10-producing CD8+ and CD4+ T cells, and occasionally FoxP3+ T regs (Wang J-P, 2010). These patients develop chronic HCV infection, while those that respond with a Th1 response clear the virus (Rouse BT, 2010). Hall et al showed that HCV-infected hepatocytes are capable of inducing T regs .…”

“…IL-10 can also inhibit pro-inflammatory cytokine signaling pathways such as NF-κB. In addition, IL-10 suppresses phosphorylation of STAT1 and induces suppressor of cytokine signaling 3 (SOCS3) expression by neutrophils and macrophages (Rouse BT, 2010). Protective immunity against hepatitis C virus is associated with a robust Th1 response and production of IFN-.…”

“…HCV successfully evades the immune response in 60% -80% of cases, resulting in chronic liver disease. Conversely, the immune response to HAV is robust and extremely effective in eliminating the virus and results in a naturally acute disease that in most cases resolves spontaneously [6,7].…”
