Immune responses to bioengineered organs

Abstract: Purpose of reviewOrgan donation in the United States registered 9079 deceased organ donors in 2015. This high percentage of donations allowed organ transplantation in 29 851 recipients. Despite increasing numbers of transplants performed in comparison with previous years, the numbers of patients that are in need for a transplant increase every year at a higher rate. This reveals that the discrepancy between the demand and availability of organs remains fundamental problem in organ transplantation.Recent findin… Show more

“…However, a major obstacle using decellularized tissue for bioengineering applications is the limited ability to recellularize the constructs and achieve an appropriate cell density which seems to play an important role in the treatment outcome. For example, recellularized constructs seem to better stimulate tissue regeneration, recruit host cells and prevent rapid scaffold degeneration compared to scaffolds without cells, in particular for larger grafts 34 37 .…”

“…Transplantation of donor tissues has been reported, but it is difficult to obtain adequate donor tissue. Even if a donor is secured and transplantation is successful, problems due to immune rejection can occur [ 4 ].…”

“…The acellular whole liver scaffold is repopulated with healthy cells to create a functional tissue that can be transplanted. One advantage of whole liver engineering approach is that the immune rejection issue can be potentially solved by engaging patient-specific liver cells such as those derived from induced pluripotent stem cells (iPSCs) in the process of recellularization 9,10 . Differentiation of iPSCs has been established for hepatocytes 1113 and more recently for cholangiocytes 1416 .…”
