Immune Responses in Vitro

Abstract: Precursors of antibody-producing cells in the mouse are functional homologues of bursa-derived lymphocytes in birds (1); they have antibody-like immunoglobulin receptors (2-8) specific for limited antigenic moieties (6-16) on their cell membrane which are largely of the 3,M immunoglobulin (Ig) 1 class in experimentally virgin animals of this species (16,17). Combination of antigen with the specific receptor and the cooperation of specific thymus-derived lymphocytes triggers division and differentiation of thes… Show more

“…Suppression has also been observed in in vitro studies (20,21). Such data strongly supports the concept that the virgin precursor "B" lymphocytes from which the humoral immune system develops express a surface receptor of~/M character.…”

“…In a further sequential ontogenetic step, ~A-bearing cells would arise from precursor cells bearing ~G receptors (12). While all studies agree on the potent immunosuppressive ability of anti-mu chain sera, experiments using anti-~ chain sera in nongerm-free states have not demonstrated an equally potent suppressor effect (20,21,23). The reason for this is not certain, but at least in vivo is probably related to the large amounts of maternal ~/G present in the circulation of newborn mice which may result in rapid removal of administered anti-),G before it can act at a cellular level (24).…”

“…It is not yet clear, however, whether IgM and IgG producers have separate precursors as opposed to a single precursor capable to switching from IgM to IgG (30,31,(35)(36)(37)(38)(39). Our observation that nonimmunized mice contain exclusively ABC with IgM receptors cannot be interpreted in favor of the latter hypothesis, because there might be separate precursors, all of which have IgM receptors, with some of them differentiating into IgM producers and others into IgG producers.…”

“…Accumulating evidence claims that B cells having/~-chain determinants on their surface are the precursors of both IgM and IgG forming cells (25,26). Such virgin B cells, termed B/z cells, would differentiate either directly into IgM-forming cells or into the cells having both /~-and ~/-chains on their surface (B/~' cells) that consequently become B~' cells, the precursors of IgG forming cells (27,28).…”
