“…The 80 chicks of 2 weeks age were divided into four groups, A, B, C and D as 20 chicks /group and then immunized by 30000, 40000 and 50000 sonicated sporulated Eimeria acervulina oocysts per os respectively and group D was left without immunization as control group. All of the four tested groups (A,B,C and D) were challenged three weeks post immunization using 50000 sporulated oocysts virulent strain/chick as recoded by (Shirly et al, 1995 andAkhtar et al, 1998), during that time fecal samples of all immunized and challenged chicks were collected daily to detect the presence or absence of oocysts, also the dead tested birds were collected for post mortem examination for recording the degree of the infection through determining the score lesions post challenge. Through this study, sonicated sporulated Eimeria acervulina oocysts were used for chicken immunization purpose. A total number of 80 susceptible chicks were divided into four groups (A, B, C and D, 20chicks/ each), the first three groups received sonicated sporulated oocysts of Eimeria acervulina in dose of 30000, 40000 and 50000 per os /one chick respectively while group D was left without immunization as a control group. The first three groups with stand challenge test using hot strain of Eimeria acervulina and showed protection percentage of 70%., 80% and 85% respectively with minimal lesion score, obvious increase body weight gain and good health condition compared with the control group which recorded high level of score lesion up to +4 with 0% protection and high mortality rate. Also there was observed reduction in fecal oocysts shedding in immunized chicks compared with the control one. On applying ELISA., it was found that there was increasing in antibody titer against Eimeria acervulina in immunized birds compared with the control group. S0 it could be corcluded that 50000 sonicated oocyct represent the best protective dose protected chicks against E.acervulina infestation.

The immunizing effect of inactivated sporulated oocyst and inactivated sonicated vaccines against bovine coccidiosis was observed in calves. Indirect haemagglutination (IHA) test was developed for detecting antibodies to coccidian. Serum antibody levels in calves were measured against soluble oocyst (sporulated) antigen. IHA antibody titer was significantly higher (P<0.05) in calves vaccinated with inactivated sonicated vaccines as compared to the calves vaccinated with inactivated sporulated vaccines. Results of the challenge experiments indicated that the inactivated sonicated vaccine gave protection to the challenge calves as immune calves contained high level of antibodies that resisted heavy dose of challenge. Disease was observed in control group (non-vaccinated) calves post challenge whereas vaccinated remained healthy. Then the oocyst count per gram of faeces was significantly higher (P<0.05) in control group (unvaccinated) as compared with the vaccinated groups.

Protective effect of egg-propagated Eimeria tenella (local isolates) gametocytes as vaccine(s) against mixed species of coccidia in chickens

Egg propagated gametocytes of Eimeria tenella (local isolates) were used to prepare the adjuvanted (Amphigen) and nonadjuvanted vaccine(s) and evaluated on the basis of cellular, humoral, and challenge responses. Modified splenic cell migration inhibition test and enzyme-linked immunosorbant assay were used to assess the cellular and humoral responses, respectively. Chicken in groups A, B, C, and D were given adjuvanted vaccine (orally), adjuvanted vaccine (subcutaneously, s/c), nonadjuvanted vaccine (orally), and nonadjuvanted vaccine s/c, respectively. Control groups E, F, G, and H were given adjuvant (orally), adjuvant (s/c), phosphate-buffered saline (PBS) + adjuvant (orally), and PBS + adjuvant (s/c), respectively. On 5 and 15 days post vaccination after boosting, significantly higher (P<0.05) cell-mediated and humoral responses were detected in vaccinated chicken compared to control. No significant effect of adjuvant and vaccination route on the immune responses was found. Maximum percent protection (survivors after challenge) against mixed species of genus Eimeria was observed in group A (71.42%) followed by group C (63.63%), B (59.09%), and group D (54.54). Significantly higher (P<0.05) oocysts per gram (OPG) of droppings was observed in the control groups compared to the vaccinated chickens. Maximum percent reduction in OPG was also recorded in group A (86) followed by group C (84), group B (83), and group D (82). From these results, it was concluded that egg-propagated gametocytes (E. tenella) that gave protection upon challenge may be due to the control of E. tenella. Further studies on its feasibility as commercial vaccine are underway.
