Immune Landscape of Thyroid Cancers: New Insights

Abstract: Immune system plays a key role in cancer prevention as well as in its initiation and progression. During multistep development of tumors, cells must acquire the capability to evade immune destruction. Both in vitro and in vivo studies showed that thyroid tumor cells can avoid immune response by promoting an immunosuppressive microenvironment. The recruitment of immunosuppressive cells such as TAMs (tumor-associated macrophages), TAMCs (tumor-associated mast cells), MDSC (myeloid-derived suppressor cells), TANs… Show more

“…1. One of the evidences of the immune evasion skills of the rumor cell is the activation of the immune checkpoint (PD-1/ PD-L1 axis, CTLA-4, TIGIT, TIM3) (Menicali et al 2020). The PD-1/PD-L1 axis activation appears to be one of the most important mechanisms of thyroid carcinogenesis (Cunha et al 2013b).…”

“…ATCs have the highest density of TAMS in their TME, which correlates with poor survival rates [ 120 ], although the importance of CAFs on thyroid cancer progression must not be forgotten [ 121 ]. The inclusion of this very important population of immunosuppressive, proangiogenic macrophages by ATC fits in well with the general concept of a locally healing permanent inflammation [ 78 ].…”

“…TC cells can maintain growth by secreting a variety of cytokines and also by secreting chemokines to recruit different immune cells ( Lumachi et al, 2010 ; Fozzatti and Cheng, 2020 ). These cytokines or chemokines affect the progression of TC and changes in the immune microenvironment ( Menicali et al, 2020 ). Innate immune cells, especially natural-killer cells (NK cells) and macrophages play a key role in the regulation of tumor progression and inhibition ( Goldberg and Sondel, 2015 ).…”
