Immortalized Epididymal Cell Lines From Transgenic Mice Overexpressing Temperature‐Sensitive Simian Virus 40 Large T‐Antigen Gene

et al.

Abstract: Epididymal epithelium is well known as a site of secretion of various proteins present in epididymal luminal fluid. Although there have been many reports of primary cultures of epididymal epithelial cells, their growth is limited over time. We have established immortalized epididymal epithelial cell lines from primary cultures of epididymal cells from transgenic mice harboring temperature‐sensitive simian virus 40 large T‐antigen gene in order to study the regulatory mechanisms of epididymal function, includin… Show more

“…Currently, culturing techniques for epididymal cells had become well-established ( Klinefelter et al, 1982 ; Chen et al, 1998 ; Lee et al, 2007 ). A rat caput epididymal cell line had demonstrated characteristics similar to epithelial principal cells found in vivo ( Araki et al, 2002 ; Dufresne et al, 2005 ). The advent of RNA sequencing technologies had enabled researchers to analyze the transcriptomes of specific groups or even individual cells ( Vazquez et al, 2019 ).…”

“…Our lab and others have developed human (Dube, Dufresne, et al., 2010; Dube, Hermo, et al., 2010), rat (Dufresne et al., 2005), and mouse (Araki et al., 2002; Sipila et al., 2004) epithelial principal cell lines, which have been used to address certain questions, such as about the regulation of junctional proteins in the epididymis (Adam & Cyr, 2016; Dufresne & Cyr, 2007; Gregory & Cyr, 2019), as well as the toxicity of nonylphenols (Jones & Cyr, 2011). Given that the epididymal epithelium consists of multiple cell types, however, a 3D in vitro model provides a more comprehensive and realistic tool that can be used to study and elucidate the multiple aspects of epididymal function.…”
