Immobilization of Interfacial Activated Candida rugosa Lipase Onto Magnetic Chitosan Using Dialdehyde Cellulose as Cross-Linking Agent

Abstract: Candidarugosa lipase (CRL) was activated with surfactants (sodium dodecyl sulfate [SDS]) and covalently immobilized onto a nanocomposite (Fe3O4-CS-DAC) fabricated by combining magnetic nanoparticles Fe3O4 with chitosan (CS) using polysaccharide macromolecule dialdehyde cellulose (DAC) as the cross-linking agent. Fourier transform infrared spectroscopy, transmission electron microscope, thermogravimetric analysis, and X-ray diffraction characterizations confirmed that the organic–inorganic nanocomposite support… Show more

“…3(a), the highest yield of 96.97% can be obtained at an optimal reaction temperature of 45 °C. Moreover, it can be seen that the optimum reaction temperature is increased to 45 °C, which is significantly higher than that of free CRL (40 °C) 44 . The temperature required for the reaction has also been reduced by 20% compared to previous work in the laboratory 10 .…”

“…2(b), compared with the free lipases, the proportion of alpha-helix of the modified lipases was significantly decreased (see Supporting information, Table S8), whereas the proportion of beta-sheet was significantly increased, which may be a satisfactory explanation for the enhancement of stability of lipases because beta-sheet is more stable than alpha-helix. 43,44 Effect of reaction medium on the synthesis of VES using CRL-IL-2…”

“…At present, the main methods to synthesize fatty acid esters of phytosterols are chemical and enzymatic. Although the enzymatic synthesis of phytosterol fatty acid esters has the advantages of mild conditions and green environmental protection, its industrial application is limited by low efficiency and high cost [ 14 , 15 , 16 , 17 ]. Therefore, chemical synthesis is still the most widely used method for the preparation of phytosterol esters.…”

“…A common chemical method to immobilize enzymes is to use glutaraldehyde [25][26][27] to connect the carrier with the enzyme molecule. 28,29 Nevertheless, the immobilization procedure involves toxic solvents, resulting in a reduction in enzyme activity. 30 Therefore, a simple, nontoxic, covalent immobilization process is crucial to solve this problem.…”
