Imbalance Between Th17 Cells and Regulatory T Cells During Monophasic Experimental Autoimmune Uveitis

Abstract: The aim of this study is to explore the dynamic changes in IL-17-expressing T cells (Th17)/Treg expression in monophasic experimental autoimmune uveitis (mEAU). mEAU was induced in Lewis rats with IRBP1177-1191 peptide and evaluated clinically and pathologically on days 9, 13, 18, 23, 28, 35, and 48. Lymphocytes isolated from inguinal lymph nodes were subjected to flow cytometry to analyze the frequency of Th17/Treg cells. The levels of cytokines (IL-17, IL-6, IL-10, transforming growth factor (TGF)-β) in seru… Show more

“…Recent reports also suggest that its breakdown may play a key role in the pathogenesis of pars planitis, which is an idiopathic chronic intermediate uveitis [ 18 ]. Several immune alterations were previously described in experimental models of uveitis [ 20 , 21 ]. According to our knowledge, there is still a lack of reports on the immune status of pars planitis patients, and the present work is the first study performed in humans.…”

“…All samples were analyzed using an Applied Biosystems ABI 7500 Real-time PCR (USA) system. PCR conditions for gene amplification were as follows: 2 min at 50 °C, followed by 40 cycles of 95 °C for 2 min, 95 °C for 15 s, and 60 °C for 30 s) [ 18 ]. The PCR products were analyzed by using a basic relative quantification method, and the amplification products were confirmed based on the observation of a single peak in the melting curve.…”

“…The imbalance ratio of Treg and helper T lymphocytes 17 (Th17) will affect the progress of interactions and is related to the occurrence and development of sepsis 21 and other diseases 22,23 .The results of flow cytometry manifested a high ratio of Th17 /Treg in sepsis murine. Both ETDCs and miR155 -/-ETDCs infusion displayed a ability to attenuate Th17/Treg ratio while the Th17/Treg ratio was lowest in the miR155 -/-ETDCs treatment group.…”
