Imbalance between matrix metalloproteinase‐1 (MMP‐1) and tissue inhibitor of metalloproteinase‐1 (TIMP‐1) contributes to bladder compliance changes in rabbits with partial bladder outlet obstruction (PBOO)

Abstract: What's known on the subject? and What does the study add?• A decrease in bladder compliance is known to be correlated with deterioration of renal function after bladder outlet obstruction (BOO). Increased deposition of extracellular matrix (ECM) in the detrusor layer is the primary reason for decreased compliance. In the bladder, as in other organs, ECM deposition is dependent on the balanced activity of proteolytic enzymes, including matrix metalloproteinases (MMPs) and their endogenous inhibitors, tissue inh… Show more

“…All of these genes have been reported to have key roles in PBOO induced bladder pathogenesis. 5,7,8,10,20 Treatment with 17-DMAG decreased PBOO induced up-regulation of the Tgfb, Glut1, Flt-1 and Timp-1 genes whereas the expression level of Vegf was similar to that in the placebo group (2.8-fold vs 2.2-fold increase vs the Sham plus P group). Many stimuli, including growth factors, hormones and cytokines, regulate Vegf expression at the transcriptional level.…”

“…In the bladder, as in other organs, the ratio of MMPs and TIMPs is critical in the regulation of ECM turnover. During BOO, the imbalance between MMP1 and TIMP‐1 was found to be associated with the accumulation of extracellular collagen and decreased bladder compliance 20 . However, to date, there is no further study stepping in the causes of this imbalance.…”

“…Negative controls were realized by omission of the primary antibody. Primary antibodies have been confirmed to cross-react with rabbit tissue [4244]. Subsequently the sections were washed with TBS and treated for 30 minutes at room temperature either with undiluted EnVision+ HRP complex (Dako), or with mouse anti-goat biotinylated secondary antibody (sc-53799, Santa Cruz; 1:200), followed by Vectastain ABC kit reagents according to the manufacturer’s instructions (Vector Laboratories, Burlingame, CA).…”
