Imaging macrophage distribution and density in mammary tumors and lung metastases using fluorine‐19 MRI cell tracking

Abstract: This study shows for the first time proof of the ability to use MRI cell tracking to visualize MAMs in the lungs. The ability to detect and monitor the number of TAMs in individual tumors with F MRI would allow for identification of breast tumors with heavy infiltration of TAMs and could be used as a biomarker for decisions about how to best treat these patients as well as for monitoring responses to therapy. “…Dual-contrast agents for 19 F-MRI and fluorescence, e.g. perfluorocarbon-TexasRed, have been used to track tumour-associated macrophages in mice ( Makela and Foster, 2018 ). MRI reporter genes have also been developed ( Table 1 ) and have the advantage of co-registration with soft-tissue anatomy and certain functional imaging parameters.…”

“…These types of labelling agents include fluorinated emulsions (CS-1000 and VS-1000H) and other fluorescently tagged formulations (CS-ATM DM Red, Green, NIR) for ex vivo and in situ cell labels. 133 , 134 A recent study showed that it is possible to effectively label hMSCs with cell sense without affecting cell viability, function or differentiation. The bSSFP sequence was used to detect and quantify the signal of Cell Sense-labeled hMSC cells in vitro and in intramuscular implantation.…”

“…Several groups, Khurana et al [120], Makela et al [121] and Huyn Shin et al [122], have also reported 19 F-based MRI cell tracking by using in vivo PFC-labelled nanoparticles to detect, quantify and track TAMs inside murine orthotopic breast cancer tumors and its associated metastasis or in mouse model of head and neck squamous cells carcinoma.…”
