Illuminating Spatiotemporal Regulation of AMPK with a Genetically Encoded Excitation‐Ratiometric Biosensor for AMPK

Abstract: AMP activated protein kinase (AMPK) is a heterotrimeric protein responsible for moderating cellular energy homeostasis. In cancer, AMPK has been implicated as a context‐dependent tumor promoter or suppressor. To illuminate spatiotemporal regulation of AMPK, which is critical for context dependent roles of AMPK, we developed a single‐fluorophore excitation‐ratiometric AMPK activity reporter (ExRai AMPKAR), which exhibits greater sensitivity and dynamic range compared to our previous AMPK biosensor. We used phar… Show more

“…Besides the well-known upstream and downstream signaling pathways, AMPK is also regulated precisely in terms of its intracellular distribution and activation 30 32 . For instance, the localization of AMPK is known to be regulated by myristoylation of the β subunit which facilitates the association of AMPK with membrane structures and the activation of AMPK by LKB1 31 , 33 , 34 .…”
