Ileal expression of gastrin and cholecystokinin

Abstract: Antibodies against the common active site of cholecystokinin (CCK) and gastrin stain three endocrine celltypes in the gut: G-cells (that synthesize gastrin) I-cells (that synthesize CCK), and TG-cells (whose product is unknown). In order to examine whether TG-cells either process progastrin or proCCK in a specific manner, or express a novel gastrin-CCK related hormone, we studied the distal porcine ileum, which have far more TGthan G-and I-cells. Ileal CCK and gastrin mRNA corresponded to those of the antroduo… Show more

“…Our transfection studies indicate that relatively profound changes in charge distribution and structure around the sulfation site do not affect sulfation dramatically. Nevertheless, eutopically expressed gastrin is completely nonsulfated in pituitary corticotrophs (42,43) and in ileal mucosa cells (44). Moreover, antral gastrin is not sulfated either in the horse 2 or in the structurally more remote turtle gastrin (37), although both proteins are readily sulfated in HIT cells.…”

“…The stomachs from overnight-fasted and anesthetized mice were collected, and whole-stomach gastrin was extracted as previously described (54). The plasma (20 μL) and stomach-extracted gastrins were measured by radioimmunoassay (EuroDiagnostic AB).…”

“…(Hortin et al, 1986;Huttner and Baeuerle, 1988;Niehrs et al, 1994 (Andersen, 1985). Complete lack of sulfation is found in the adenohypophysis (Rehfeld and Larsson, 1981) and in the ileum (Friis-Hansen and Rehfeld, 1994). In contrast, gastrin is completely sulfated in the fetal pancreas (Brand et al, 1984).…”

“…Additional functions in progastrin maturation have been suggested earlier. Thus, examinations of plasma and tissue samples from hypergastrinaemic patients showed a direct correlation between the degree of sulfation and the maturation of progastrin to gastrin-17 (Andersen and Stadil, 1983;Jensen et al, 1989 Rehfeld, 1994).…”
