Ile-177 and Ser-180 in the S1 Segment Are Critically Important in Kv1.1 Channel Function

Abstract: Ile-177 and Ser-180 are conserved residues in the first transmembrane segment (S1) of the Shaker, Shab, Shaw, and Shal subfamilies of voltage-gated K ؉ channels. Here we report that the mutation of these residues in Kv1.1 to leucine, proline, or arginine abolished the expression of outward potassium currents in Xenopus oocytes. Co-injection of these mutant cRNAs and wild type Kv1.1 cRNA into Xenopus oocytes exerted a potent dominant negative effect resulting in the suppression of Kv1.1-encoded currents. Transi… Show more

“…PalmϪ stable CHO cells were grown to 50 -60% confluence on glass coverslips in six-well plates, fixed in 4% paraformaldehyde in PBS for 15 min at room temperature, permeabilized with 0.2% Triton X-100 in PBS for 5 min at room temperature, and blocked with 5% donkey serum in PBS for 1 h at room temperature (29,31). The cells were then incubated with rabbit polyclonal or mouse monoclonal antibodies at 1:100 -250 in 5% donkey serum for 1 to 2 h at room temperature.…”

“…The importance of Ile177 for Kv1.1 channel function has been inferred from the expression characteristics of several mutations of this residue [21]. However, this study did not report biophysical evidence supporting this conclusion, as the mutations analysed abolished channel function.…”

“…No gating currents could be detected in 15 out of 114 mutants (minus signs "-" in Table 1). Among them, the homologous mutations I237R, S240L, S240P, and S240R in the related potassium channel Kv1.1 were previously shown to abolish K + conduction (49).…”
