IL6-STAT3-HIF Signaling and Therapeutic Response to the Angiogenesis Inhibitor Sunitinib in Ovarian Clear Cell Cancer

Abstract: Purpose: Ovarian clear cell adenocarcinoma (OCCA) is an uncommon histotype that is generally refractory to platinum-based chemotherapy. We analyze here the most comprehensive gene expression and copy number data sets, to date, to identify potential therapeutic targets of OCCA.Experimental Design: Gene expression and DNA copy number were carried out using primary human OCCA tumor samples, and findings were confirmed by immunohistochemistry on tissue microarrays. Circulating interleukin (IL) 6 levels were measur… Show more

“…Further investigation documented that cucurbitacin I treatment inhibited the JAK2/STAT3 pathway, accompanied by decreased levels of HIF-1␣, which echoed the findings that the JAK2/STAT3 pathway positive regulates HIF-1␣ in a variety of cancer cell types (37)(38)(39)(40). HIF-1␣ plays an important role in hypoxia-induced autophagy, requiring specific activation of an autophagy-inducing molecule, BNIP3, which is activated by HIF-1 only in hypoxic cells (53).…”

“…However, a recent study performed by Panici and colleagues suggest that the survival between the two groups to be far closer (72 months vs 62 for types I and II respectively [6]), indicating that there is still much we don't understand about this disease. Adding to this argument, is the finding that clear cell ovarian cancer (CCOC) is genetically separate to other forms of ovarian cancer, with different common mutations [7], and de novo chemo-resistance.…”

“…Ongoing molecular characterisation and genetic studies suggests that this model may be overly simplistic, with a number of subtypes being re-classified, or further divided into additional sub-groups [8,9,7,10]. These new subtypes demonstrate different genetic lesions and transcriptional profiles, chemo-sensitivity, and survival rates, indicating a need to incorporate this information into treatment plans for ovarian cancer.…”

“…Many of the earlier classification studies in ovarian cancer focused on defining gene signatures of known traditional histologic ovarian cancer subtypes including the rare histologic types such as clear cell, mucinous or endometrioid ovarian cancer. These analyses show that clear cell and mucinous ovarian cancers can be readily distinguished from serous ovarian cancers based on their gene expression profiles, regardless of tumor stage and grade [33][34][35][36]. One of these studies also confirms that low-grade endometrioid cancers are distinctly separated from the other groups of ovarian tumors [37].…”
