IL2RG, identified as overexpressed by RNA-seq profiling of pancreatic intraepithelial neoplasia, mediates pancreatic cancer growth

Abstract: Pancreatic ductal adenocarcinoma evolves from precursor lesions, the most common of which is pancreatic intraepithelial neoplasia (PanIN). We performed RNA-sequencing analysis of laser capture microdissected PanINs and normal pancreatic duct cells to identify differentially expressed genes between PanINs and normal pancreatic duct, and between low-grade and high-grade PanINs. One of the most highly overexpressed transcripts identified in PanIN is interleukin-2 receptor subunit gamma (IL2RG) encoding the common… Show more

“…S2D-S2F). Decreased tumor burden aligns with previous reports that shRNA-mediated IL2Rγ depletion provokes tumor regression (17). These modest antitumor results raised the possibility that either IL2Rγ plays a marginal role in tumor growth, or an alternate receptor complex or pathway is available to sustain growth and survival signals.…”

“…In human pancreatic intraepithelial neoplasia-3 (PanIN-3), IL2RG was shown to be highly expressed. Knockout by CRISPR/Cas9 of IL2RG of bkpc58 cells derived from KPC mice attenuated tumor growth in an orthotopic mouse model of PDA via the Jak3 pathway 25 . However, KPC mice do not express CEACAM6 and a functional correlation to IL-2RG cannot be rationalized.…”

“…In tissue samples from patients with SD or PD, IL2RG (IL-2 receptor subunit gamma) was differentially expressed after treatment, which was previously identified to contribute in pancreatic cancer growth [ 65 ]. In post-therapy samples, increased expression of SLAMF7 , CD38 , C7 , and CD79A was associated with lack of radiologic response ( S4A Fig ), and increased expression of these genes was also associated with pathologic non-responders ( Fig 5C ).…”
