IL23R in the Swedish, Finnish, Hungarian and Italian populations: association with IBD and psoriasis, and linkage to celiac disease

Abstract: BackgroundAssociation of the interleukin-23 receptor (IL23R) with inflammatory bowel disease (IBD) has been confirmed in several populations. IL23R also associates with psoriasis, suggesting that the gene may be an important candidate for many chronic inflammatory diseases.MethodsWe studied association of single-nucleotide variants in IL23R with IBD in Swedish patients, in both Crohn's disease (CD) and ulcerative colitis (UC) subsets. The same genetic variants were also studied in Finnish patients with psorias… Show more

“…By contrast, multiple genes implicated in the IL-23 pathway, including IL23R (encoding a subunit for IL-23 receptor) and IL12B (encoding the p40 subunit of IL-12 and IL-23), STAT3 (encoding signal transducer and activator of transcription 3) and the homeodomain-containing transcription factor NKX2-3 have been shown to be associated with both UC and CD. 25,[29][30][31][32][33] On the other hand, mutations in ECM1 (extracellular matrix protein 1) are specific to patients with UC. 21,31,32 Microbiota in IBD.…”

“…The genes IL12B and IL23R are involved in the regulation of this pathway in Crohn's disease and ulcerative colitis. A study of the Swedish and Finnish population confirms the association between psoriasis and inflammatory bowel diseases, in which both are associated with the IL23-R gene 10 . A highly significant relationship between Crohn's disease and the gene for IL23-R localized on chromosome 1p31 was confirmed by genomic association studies performed in patients with Crohn's disease 11,12 .…”

“…Although IL-23 has its own unique subunit, IL-23R, together with IL12Rb1, forms the signaling complex [22,23]. IL-23R gene becomes one of the most talked about candidate genes in recent years, and its susceptibility alleles associated with inXammatory bowel disease [24,25], psoriasis [25], and AS [11,12]. The IL-23R gene is located on chromosome 1p31, lie on the region of susceptibility genes obtained by linkage analysis.…”
