IL-9 Inhibits Oxidative Burst and TNF-α Release in Lipopolysaccharide-Stimulated Human Monocytes Through TGF-β

Abstract: IL-9 is a Th2 cytokine that exerts pleiotropic activities on T cells, B cells, mast cells, hematopoietic progenitors, and lung epithelial cells, but no effect of this cytokine has been reported so far on mononuclear phagocytes. Human blood monocytes preincubated with IL-9 for 24 h before LPS or PMA stimulation exhibited a decreased oxidative burst, even in the presence of IFN-γ. The inhibitory effect of IL-9 was specifically abolished by anti-hIL-9R mAb, and the presence of IL-9 receptors was demonstrated on h… Show more

“…Because it is known that monocytes of sepsis patients often show a desensitized phenotype (6,20), we elucidated the underlying mechanism. An attenuated respiratory burst in monocytes/macrophages has been described in response to IL-4, causing LPS-receptor (CD14) and TLR-4 downregulation (24). These transcriptional mechanisms prevent signal transmission provoking cellular activation following LPS binding.…”

“…Human IL-9 was purified as previously described [7]. LPS from Escherichia coli (serotype O55:B5) was purchased from Difco Laboratories (Detroit, MI, USA), and anti-TGF-b1 mAb (clone TB21, mIgG1) was from Biosource International (Camarillo, CA).…”

“…CD14 was detected at the surface of AM, although to a much less extent than on blood monocytes [7]. CD14 expression at the surface of AM was downregulated by preincubation with IL-4 (mean fluorescence intensity (MFI) 42¡4 versus 84¡6, pv0.001).…”

“…IL9r was shown to inhibit both the production of reactive oxygen species and the release of TNF-α by LPS-stimulated human monocytes. 35 Via toll-like receptor 3, 36 stathmin is thought to mediate IL-10 induction, 37 which is increased in the plasma of apoE…”
