IL-37 suppresses hepatocellular carcinoma growth by converting pSmad3 signaling from JNK/pSmad3L/c-Myc oncogenic signaling to pSmad3C/P21 tumor-suppressive signaling

Abstract: IL-37 has been characterized as a fundamental inhibitor of innate immunity and a tumor suppressor in several cancers. However, the molecular mechanism of IL-37 in hepatocellular carcinoma (HCC) is largely unclear. In this study we found IL-37 expression was down-regulated in human HCC tissues and cell lines, and was negatively correlated with tumor size, vascular invasion, as well as overall-survial and disease-free survival (OS and DFS) of HCC. Multivariate Cox analysis revealed that IL-37 was an independent … Show more

“…Current studies indicate that, many cytokines, including IL‐17, IL‐6 and IL‐37, could regulate the function of HCC cells by an autocrine manner . We next examined whether HBX could promote the secretion of IL‐34 to mediate the biological function of HCC cells.…”

“…Furthermore, when knocking down of IL‐34 by shRNA, the proliferation and migration efficiency of HCC cells mediated by HBX was decreased. Previous studies show that multiple autocrine cytokines, including IL‐17 and IL‐37 , could regulate the biological function of hepatoma cells . We next investigated whether HBX could promote the secretion of IL‐34 from HCC cells and the secreted IL‐34 in turn had an effect on HCC cells.…”

“…Acting as a dual‐function cytokine, intracellular IL‐37 can regulate cellular behavior. Overexpression of IL‐37 in human hepatocellular carcinoma (HCC) cell lines HepG2 and SMMC‐7721 significantly suppressed HCC cell growth and proliferation through converting Smad3 phospho‐isoform . Intracellular mature IL‐37 can directly bind to Rac1 and subsequently suppress the migration ability of A549 and BEL‐7402 cell .…”
