IL-18BP is decreased in osteoporotic women: Prevents Inflammasome mediated IL-18 activation and reduces Th17 differentiation

Abstract: IL-18BP is a natural antagonist of pro-inflammatory IL-18 cytokine linked to autoimmune disorders like rheumatoid arthritis. However, its role in post menopausal osteoporosis is still unknown. In this study, we investigated the role of IL-18BP on murine osteoblasts, its effect on osteoblasts-CD4+ T cells and osteoblasts-CD11b+ macrophage co-culture. mIL-18BPd enhances osteoblast differentiation and inhibits the activation of NLRP3 inflammasome and caspase-1 which process IL-18 to its active form. Using estroge… Show more

“…Micro-computed Tomography-CT determination of excised bones was carried out using Sky Scan 1076 CT scanner (Aartselaar, Belgium) using previously published protocol (10,24,36). Three samples were scanned per batch at nominal resolution pixel 18 m. Reconstruction was carried out using a modified Feldkamp algorithm using the Sky Scan Nrecon Software, which facilitates network-distributed reconstruction carried on a personal computer running simultaneously.…”

“…In addition, signals provided by NF- κ B activators are necessary for NLRP3 inflammasome activation to produce active IL-1 β and IL-18 [ 31 ]. The NLRP3 inflammasome also potentially creates a microenvironment for Th17 cell differentiation [ 38 , 39 ]. Thus, the NLRP3 inflammasome regulated by the NF- κ B pathway, at least partially, may take part in ITP pathogenesis mediated by the dysfunction of T helper cells.…”

“…In post-menopausal osteoporosis, IL-18 blood levels are increased while those of its antagonist, IL-18BP, are decreased. According to Mansoori et al, IL-18BP enhanced murine osteoblast differentiation and inhibits the activation of NLRP3 inflammasome and caspase-1, in vitro , and improved the metabolic and bone statuses in ovariectomized rats (a rodent model of post-menopausal OP) (132). Further evidences derived from the association of NLRP3 mutations with arthropathy and OP (133) and the SIRT1-dependent inhibition of osteogenic differentiation and enhancement adipogenic differentiation, in mesenchymal stem cells (MSC), following lipopolysaccharide (LPS)-induced NLRP3 inflammasome activation (134).…”
