IL-17 in osteoarthritis: A narrative review

Juan Xiao,
Ping Zhang,
Fang-Lan Cai
et al.

Abstract: Osteoarthritis (OA) is a painful joint disease that is common among the middle-aged and elderly populations, with an increasing prevalence. Therapeutic options for OA are limited, and the pathogenic mechanism of OA remains unclear. The roles of cytokines and signaling pathways in the development of OA is a current research hot spot. Interleukin (IL)-17 is a pleiotropic inflammatory cytokine produced mainly by T helper 17 cells that has established roles in host defense, tissue repair, lymphoid tissue metabolis… Show more

“…The results of KEGG analysis showed that RJNTF could treat KOA by interfering with multiple signaling pathways such as NF-κB, apoptosis, HIF-1 signaling pathway, JAT-STAT signaling pathway and IL-17 signaling pathway ( Fig. 2B ) ( 31 34 ). Based on the results of a large number of previous studies showing that the PARP1/AIF pathway was closely related to apoptosis ( 35 37 ), while the results of network pharmacology indicated that the treatment of KOA by RJNTF was related to the PARP1 apoptotic pathway; however, there are no studies assessing the relationship between the PARP1/AIF pathway and apoptosis of chondrocytes.…”

“…The IL-17 signaling pathway has been repeatedly reported in relation to OA. The IL-17 family consists of six cytokines (IL-17A to F), with IL-17A being the most extensively studied and considered the most influential [ 15 , 17 , 31 , 32 ]. Studies have shown that the levels of IL-17A in the serum and synovial fluid of OA patients are higher than those in healthy individuals and are positively correlated with measures of pain, functionality, and disease severity [ 18 , 33 ].…”

“…The IL-17 signaling pathway has been repeatedly reported to be related to OA. IL-17 is a family of six cytokines (IL-17A-F), with IL-17A being the most studied and significant [ 17 , 31 , 32 , 42 ]. Studies have shown that the extracellular matrix (ECM) regulates the activation state of cells by blocking or facilitating the binding of cytokines to cell surface receptors.…”
