IL-10 reduces apoptosis and extracellular matrix degradation after injurious compression of mature articular cartilage

Abstract: Joint injury is a complex process involving specific mechanical effects on cartilage as well as induction of an inflammatory environment. IL-10 prevented crucial mechanisms of chondrodegeneration induced by an injurious single compression. IL-10 might be a multipurpose drug candidate for the treatment of cartilage-related sports injuries or osteoarthritis (OA).

“…Many differences between OA and PTOA are known; we refer to the PTOA literature [234,235]. The here discussed data have not been derived from studies that had PTOA in mind; e.g., models of mechanical overload such as those described in [236,237], or studied therapeutic options after AC injury [238,239,240]. Thus, it remains unclear whether the here derived insights are valid under conditions known to lead to PTOA and designated studies using standardized injury models are needed.…”

“…For applying compressive overload, an incubator‐housed loading device capable of axial compression was used similar to previous studies (Behrendt et al, ; Rolauffs et al, ). When placing the specimens into the loading chamber, their individual thickness was measured by applying a slow compression 20 μm/s ramp speed with a manual compression interrupt when a sudden increase in offset load was noted that indicated contact between platen and disk.…”

“…Among these enzymes, MMP‐13 predominantly cleaves COL II . ADAMTS5 is responsible for proteoglycans degradation, and NOS2 induced NO production which attribute to apoptosis of chondrocytes . Our results showed that FS of S. aureus induced expression of ADAMTS5, NOS2, MMP3, MMP13 in mature differentiated chondrocytes at passage2.…”
