IL-1.BETA. Genotype-Related Effect of Prednisolone on IL-1.BETA. Production in Human Peripheral Blood Mononuclear Cells under Acute Inflammation

Abstract: Glucocorticoids are potent anti-inflammatory and immunosuppressive drugs and consequently are widely used in the treatment of systemic inflammatory, allergic, and autoimmune diseases. The anti-inflammatory properties of glucocorticoids are mainly ascribed to their ability to suppress the synthesis of pro-inflammatory cytokines.1) But there is a certain percentage of patients, resistant to glucocorticoid-based anti-inflammatory therapy, who show high levels of local and/or systemic pro-inflammatory cytokines du… Show more

“…IL-1β expression and markers of classical macrophage activation have been observed in bronchoalveolar fluid lavage of patients with steroid-resistant asthma (45). IL-1β production in response to prednisolone, in part, may be modulated by IL-1β genotype, as follows: patients with certain IL-1β genotypes experience reduced responses to prednisolone (46). Genetic studies of single-nucleotide polymorphisms in IL-1β and IL-1β family members are in progress.…”

“…Further, our UC patients were all corticosteroid naive for the current flare-up, as corticosteroids are reported to suppress the levels of cytokine mRNA. 25 With these 3 groups we investigated the levels of cytokine mRNAs in the mucosal tissue at the inflammatory sites, 23 and appar-FIGURE 2. “…IL-1B genotypes at positions -511, -31 and 3954 were determined by polymerase chain reaction (PCR)-restriction fragment length polymorphism assays as described earlier [43,44,49], except that different forward and reverse primers were used for the 3954 assay with the following sequences: forward, 5 0 -CTC AGG TGT CCT CCA AGA AAT CA-3 0 ; reverse, 5 0 -GCT TTT TTG CTG TGA GTC CCG-3 0 .…”
