IgG subclasses of anti‐FVIII antibodies during immune tolerance induction in patients with hemophilia A

Abstract: SummaryThe eradication of inhibitory antibodies in patients with haemophilia A can be accomplished by frequent administration of high or intermediate doses of factor VIII (FVIII), so-called immune tolerance induction (ITI). This study monitored the distribution of IgG subclasses of anti-FVIII antibodies during ITI. FVIII-specific antibodies of subclass IgG1 were detected in all inhibitor patients tested, anti-FVIII IgG4 in 16, IgG2 in 10 and IgG3 in one of 20 patients analysed. Levels of anti-FVIII IgG1 and Ig… Show more

“…The results are shown in Table 1 for 25 subjects from CHU Sainte-Justine (subjects 1-25) and Table 2 for 7 subjects [26][27][28][29][30][31][32]. fVIII antigen concentration and activity were determined as previously reported.…”

“…Immunoassays have been used to detect anti-fVIII antibodies, but report only the presence of antibody relative to controls or quantitation based on recombinant constructs as calibrators. [27][28][29][30][31][32][33][34] An assay for the detection of such antibodies with a suitable calibrator for absolute quantitation has been unavailable.…”

“…IgG 1 and IgG 4 also predominated in patients with inhibitors in acquired and in congenital hemophilia A [15], in blistering skin diseases [16], and in other autoimmune diseases [17]. Interestingly, and inexplicably, absolute levels of IgG 1 and IgG 4 were always inversely correlated.…”
