IGFBP‐3 inhibits Wnt signaling in metastatic melanoma cells

Abstract: In previous works, we have shown that insulin‐like growth factor‐binding protein‐3 (IGFBP‐3), a tissue and circulating protein able to bind to IGFs, decreases drastically in the blood serum of patients with diffuse metastatic melanoma. In agreement with the clinical data, recombinant IGFBP‐3 was found to inhibit the motility and invasiveness of cultured metastatic melanoma cells and to prevent growth of grafted melanomas in mice. The present work was aimed at identifying the signal transduction pathways underl… Show more

“…However, only the expression of p-GSK-3 β was decreased along with ILK in a dose-dependent manner, while p-Akt failed to participate in the process. It is well known that decrease of the expression of p-GSK-3 β led to the increase of GSK-3 β kinase activity, followed by phosphorylation of β -catenin [39, 40]. Similarly, our present results showed downregulated β -catenin after treatment with emodin.…”

“…Similarly, the activity of SERPINE2 stimulates the melanoma metastatic behaviour [ 73 ]. On the other hand, the role of the IGFBP3 gene in cancer is not entirely understood, because both melanoma-supporting and inhibiting effect was shown concerning this factor [ 74 , 75 ]. The activity of genes encoding E- and N-cadherins ( CDH1 , CDH2 ) was detected in melanoma cells prepared from the spheroids.…”

“…Glycosylated recombinant His-tagged IGFBP-3 (Speed BioSystems) and nonglycosylated recombinant IGFBP-3 (MyBioSource, San Diego, CA, USA) were used as reported earlier [69]. Glycosylated human CD44 recombinant protein was purchased from Sino Biological (Wayne, PA, USA).…”
