IGF2BP3 From Physiology to Cancer: Novel Discoveries, Unsolved Issues, and Future Perspectives

Abstract: RNA network control is a key aspect of proper cellular homeostasis. In this context, RNA-binding proteins (RBPs) play a major role as regulators of the RNA life cycle due to their capability to bind to RNA sequences and precisely direct nuclear export, translation/degradation rates, and the intracellular localization of their target transcripts. Alterations in RBP expression or functions result in aberrant RNA translation and may drive the emergence and progression of several pathological conditions, including… Show more

“…In fact, IGF2BP3 might differentiate normal tissues from cancerous tissues and serve as a prognostic marker for colorectal, hepatocellular, and ovarian clear-cell carcinomas [25][26][27]. Previous research has con rmed that IGF2BP3 is involved in cell growth and migration in early embryonic development [28]. Similarly, both of our results con rmed the role of IGF2BP2 and IGF2BP3 in inhibiting tumour progression.…”

“…More specifically, it has been previously shown that IGF2BP3 is upregulated across all NSCLC subtypes and that its loss leads to increased apoptosis (55). As a RBP, IGF2BP3 regulates several genes primarily by binding and stabilizing target mRNAs to increase translation efficiency (47). Two such targets, as mentioned previously, are c-Myc and HMGA2.…”

“…4). In normal tissues, IGF2BP3 is primarily expressed during embryonic development, but its overexpression has been associated with numerous cancer types and as such has been classified as an oncogenic protein (47,54). More specifically, it has been previously shown that IGF2BP3 is upregulated across all NSCLC subtypes and that its loss leads to increased apoptosis (55).…”

“…Their role in the cancer process is better understood. An increase in IGF2BP3 expression is associated with increased cell invasiveness and greater dynamics of the tumor process IGF2BP2, in turn, stabilizes IGF-2 mRNA, which increases its expression contributing to tumor development [133][134][135]. These examples indicate how extensive and important a role insulin-like growth factor 2 plays in the cancer process.…”
