IGF-binding protein-1 inhibits IGF effects on adipocyte function: implications for insulin-like actions at the adipocyte

Abstract: IGF action in vivo is acutely regulated by IGF-binding protein-1 (IGFBP-1) and its phosphorylation state is implicated in modulating these effects. Since IGFs have an important regulatory role in adipocyte function, we investigated the effects of phosphorylated IGFBP-1 (pIGFBP-1) and non-phosphorylated IGFBP-1 (npIGF BP-1) on 3T3-L1 preadipocyte proliferation and adipocyte metabolism.IGFs stimulated clonal expansion of 3T3-L1 cells (IGF-I more potently than IGF-II (EC 50 : 30 nM and 50 nM)). npIGFBP-1 inhibite… Show more

“…However, IM subjects showed a tendency towards increased fat mass and reduced adiponectin in comparison to the UM group. This finding could be consistent with a role of IGF2 in fat tissue growth and development [43,44,45,46]. …”

“…In primary rat adipocytes, Ͼ30 nM (480 ng/ml) leptin almost completely inhibits metabolic activities associated with insulin, such as stimulation of glucose transport, glycogen synthase, lipogenesis, and protein synthesis (41). Insulin is a mitogenic factor and has been shown to increase proliferation and differentiation of preadipocytes in cell lines (52,54). Leptin and insulin activate many of the same pathways, such as mitogenactivated protein kinase (MAPK), signal transducer and activator of transcription 1 and 3 (STAT1 and STAT3), and phosphatidylinositol 3-kinase (7); therefore, high concentrations of leptin could activate these pathways and interfere with insulin signaling.…”

“…IGFBP-1 concentrations are higher in women than men [17]. Phosphorylated forms of IGFBP-1 acutely inhibit IGF effects on 3T3-L1 preadipocytes and adipocytes [18]. Specific proteolysis of IGFBP-1 generates IGF-binding fragments with reduced IGF-II-stimulated glucose uptake in muscle [19].…”
