Ig-binding Receptors on Human NK Cells as Effector and Regulatory Surface Molecules

Abstract: The receptors on human natural killer 9NK cells which can specifically bind the Fc portion of immunoglobulin molecules (Fc receptors) have been extensively studied. The best known and studied Fc receptor on human NK cells is FcgammaRIIIa. Interactions of NK cells with IgG antibodies via this receptor are well known to induce a signal transduction cascade and lead to antibody-dependent cell-mediated cytotoxicity (ADCC) as well as release of various cytokines. In addition, interactions with monomeric IgG and Fcg… Show more

“…The presence of the Fc portion may have caused preferential activation of macrophages and/or NK cells, because these cells express Fc␥Rs. It is also conceivable that a negative signal may have been transmitted to the immune response through the Fc␥Rs (48). It is noteworthy that all surviving mice had high levels of specific IgM that promoted C3 deposition on Meth A cells.…”

“…Furthermore, the FcgRIIIA but not FcgRIIIB binds monomeric IgG1 and IgG3 with low affinity, in a specific and saturable manner [23][24][25], and I-mIgG for 1 h at 0 C, washed and less than 7% of the total radioactivity was counted in the culture medium. Thereafter, the cells were incubated at 37 C for different time intervals and the radioactivity in the supernatant, cell pellet or elution at pH 3.0 was measured with a Well Type Gamma Counter.…”

“…Because most Fc receptor (FcR)-bearing immune cells coexpress both activating and inhibitory FcRs that bind IgG with comparable affinity and specificity, coengagement of activating and inhibitory FcRs by IgG or immune complexes usually occurs, thereby establishing thresholds for activation/repression and determining the magnitude of effector responses (10,11). The majority of NK cells, however, do not routinely express inhibitory FcRs, and thus much less is known about inhibitory signals that regulate or attenuate NK cell effector responses following FcgRIIIa engagement (12).…”
