IFNγ Enhances Microglial Reactions to Hippocampal Axonal Degeneration

Abstract: Glial reactivity is implicated in CNS repair and regenerative responses. Microglia, the cells responding earliest to axonal injury, produce tumor necrosis factor-alpha (TNFalpha), a cytokine with both cytopathic and neuroprotective effects. We have studied activation of hippocampal microglia to produce TNFalpha in response to transection of perforant path axons in SJL/J mice. TNFalpha mRNA was produced in a transient manner, peaking at 2 d and falling again by 5 d after lesioning. This was unlike other markers… Show more

“…98,99 Using the PP lesion model, our group has previously demonstrated a transient increase in TNF mRNA expression levels at 2 days post lesion, 34 which was increased by the presence of IFN-␥. 100 This was confirmed in the present study, in which a marked increase in TNF mRNA expression was observed in T PLP mice at 2 days post lesion and persisted at 7 days post lesion. Microglial production of TNF has been previously associated with increased phagocytosis of myelin debris in microglia in vitro.…”

“…Previous studies identified microglia and astrocytes as a prominent source of CCL2 (17). IL-1b and TNF are also glial derived (37,38), and we must assume that the leukocyte contribution was not significant in this case. The differences in effect on CXCL10 may reflect involvement of adjuvant in the EAE model or other differences between these systems.…”

“…IFNg could participate in an inflammatory cascade that would also include the activation of microglial cells, as IFNg has been shown to enhance microglial cell activity in vivo. 41 Whether IFNg directly activates microglial cells or mediates astrocytic-dependent microglia activation remains to be determined. Moreover, activated microglia could participate with reactive astrocytes in the chronic production of neurotoxic molecules that could function through different mechanisms in different motoneuron populations (Figure 8).…”
