Idiotypic mimicry and the assembly of a supramolecular structure: an anti-idiotypic antibody that mimics taxol in its tubulin-microtubule interactions.

Abstract: Taxol, rii y extrced fro the bark of the western yew, Taxus brenfolia, is reportedly the first of a new class of anti-cancer agent. It Taxol, reportedly the first of a new class of anti-cancer agents, was originally extracted from the bark of the western yew, Taxus brevifolia (1). In vivo, it prevents the disassembly of microtubules, causing the arrest of cells at the G2/M step of the cell cycle (2). Its mechanism differs from that of other anti-cancer agents that have tubulin as their target, such as colchi… Show more

“…Similarities suggest that paclitaxel is a peptidomimetic compound able to mimic the endogenous ligand of the paclitaxel binding proteins. This hypothesis was proposed by Leu and colleagues (31), who discovered that a monoclonal anti-idiotypic antibody generated against paclitaxel was able to simulate paclitaxel activity, thereby showing that a protein could mimic paclitaxel. Functionally, the mimicked motif delivers a potent death signal.…”

“…40 The Paclitaxel anti-idiotypic antibody model is an example especially relevant to our work because both Paclitaxel and tobacco polyphenols share aromatic ring structure and bear no resemblance to immunoglobulin molecules. Yet both the Paclitaxel-like antiidiotypic antibody and TAIA display functional activity identical to the original antigen.…”

“…Sequencing of the variable regions of 1-10F-8A was carried out as described (20) with the following changes: total RNA was prepared using the TRIzol reagent (GIBCO͞BRL). The first strand cDNA preparations used the random hexamers of the universal riboclone cDNA synthesis system (Promega).…”
