Idiotypic and anti-idiotypic antibodies against polycyclic aromatic hydrocarbon in human blood serum are new biomarkers of lung cancer

Abstract: Evaluation of epidemiologic risk factor in relation to lung cancer invoked by polycyclic aromatic hydrocarbons has been inconsistent. To address this issue, we conducted a prospective evaluation of new biomarkers for lung cancer classified according levels of idiotypic and anti-idiotypic antibodies against polycyclic aromatic hydrocarbons in human blood serum. The blood serums of 557 lung cancer patients and 227 healthy donors were analysis of these antibodies by ELISA. Collected data were regrouped and analyz… Show more

“…In the system using a panel of 7 auto-Abs against: p53, CAGE, NY-ESO-1, GBU4-5, SOX2, MAGE A4 and Hu-D, the sensitivity was 47%, the specificity was 90%, and AUC was 0.9. Also, the new method was comparable to previously published one where Bp-BSA and mouse scFv against Bp used for detection of endogenous Ab1 and Ab2 levels against Bp in sera, which gave AUC = 0.9 at logistic regression model [16].…”

“…The proposed method as well as the previously published one [16] shows an increase in serum Abs levels in LC patients compared to healthy people. The small difference between the data presented and those previously published can be explained by the different spectrum of serum Abs that interact with the antigens absorbed onto the immunological plate or by the difference in the number of tested Interestingly, despite these minor differences, the ratio of Ab2 to Ab1 is very similar for both methods.…”

“…Biomarkers of Exposure These markers highlight the significance of various exposure pathways and cancer risk; they allow measurement of carcinogens of interest in the body from an accessible biological matrix such as blood, urine, normal, and tumor tissues. For individuals who are either occupationally or dietarily exposed to polycyclic aromatic hydrocarbons (PAHs) (risks for lung cancer [32]) or aflatoxin B (risks for liver cancer [15]) either individually or in mixtures, metabolites of these chemicals in serum or urine serve as markers of exposure.…”
