Identification of β,β-turns and unordered conformations in polypeptide chains by vacuum ultraviolet circular dichroism

Abstract: Different conformations of polypeptides were characterized by measurements of the circular dichroism (CD) extended into the vacuum ultraviolet region. (i) The linear ,-pleated sheet structure was characterized in a broad ultraviolet region down to 165 nm by examination of copolypeptides composed of alternating hydrophobic and hydrophilic aminoacid residues, e.g., poly(Lys-Leu-Lys-Leu). A short-wavelength intense band was foud at about 169 nm, which is characteristic of a-pleated sheet conformation. (ii) The #-… Show more

“…The minimum at 222 nm was red-shifted to ϳ225 nm, and the intensity of the negative band at 208 nm was significantly reduced. These new features are consistent with a substantial increase in ␤-structures in the solid fraction of the sample (43). Remarkably, solid material isolated from MPO-2:1-H 2 O 2 -apoA-I displayed similar CD spectral features (Fig.…”

“…All rED3s have similar negative ellipticity at 216 -217 nm, which is characteristic of ␤-strand structure (37) and is in agreement with the NMR structure of WNV rED3-WT (22). However, each rED3 exhibited a different spectrum at 205-207 nm that corresponds to ␤-turn structures (37,38) (Fig. 6).…”

“…The CD result should be interpreted as a specific tertiary structure, not a randomly distributed population of molecules in solution. Indeed, different distinct random conformations may display varying magnitudes of the characteristic trough at 195-197 nm; a 15% difference in ellipticity has been observed between poly(Lys+) and poly(Pro-Lys+-Leu-Lys+-Leu), both believed to be in random states (Brahms et al, 1977). Such a random coil structure is also consistent with the hydrodynamic properties of the RED (Vissavajjhala & Ross, 1990) and does not mean that the RED is unstructured.…”

“…The spectrum of RED is characterized by a slight maximum at about 225 nm and a strong minimum near 197 nm with an amplitude of -25,000 deg.cm2/dmol. This spectrum is similar to protonated polylysine in the random coil state (Brahms et al, 1977) and suggests that RED in solution exists primarily in a disordered conformation with little secondary structure. A &turn structure is the only other derived structure (Compton & Johnson, 1986) that resembles that of the RED.…”
