“…Proteins from the fermentation broth were separated by 2D gel electrophoresis [51]. Selected spots were cut from gels manually, and, after trypsin digestion, peptides were analyzed on a 4000 QTRAP (AB Sciex, Framingham, MA, USA) mass spectrometer coupled to an Easy nLC II nanoHPLC (Bruker, Billerica, MA, USA).…”

“…In S. aureus , a unique enzyme, the penicillinase encoded by the blaZ gene, is responsible for penicillin degradation. The only study of penicillinase detection by proteomic methods was performed using bovine and human isolates of borderline oxacillin‐resistant S. aureus strains . Using a gel‐based approach, the authors identified BlaZ enzymes and a longer variant with high sequence coverages of 22 to 79% in the supernatant fraction and of 18 to 82% in the membrane‐bound fraction.…”

“…However, Keseru et al. showed that the mechanism responsible for S. aureus low‐level or borderline methicillin resistance was due to the production of penicillinases similar to BlaZ . They also observed slight sequence variations in these proteins that might account for their different sizes and substrate specificity.…”

“…Another type of oxacillin-resistant S. aureus is the borderline oxacillin-resistant S. aureus (BORSA) phenotype that contains alternative resistance mechanisms. These strains include β-lactamasehyperproducing S. aureus and S. aureus with point mutations in other PBP genes [13][14][15][16][17][18]. There are no strict laboratory diagnostic criteria to define these resistance phenotypes.…”
