Identification of viral genes essential for replication of murine γ-herpesvirus 68 using signature-tagged mutagenesis

Abstract: ␥-Herpesviruses, Epstein-Barr virus, and Kaposi's sarcoma-associated herpesvirus are important human pathogens, because they are involved in tumor development. Murine ␥-herpesvirus-68 (MHV-68 or ␥HV-68) has emerged as a small animal model system for the study of ␥-herpesvirus pathogenesis and host-virus interactions. To identify the genes required for viral replication in vitro and in vivo, we generated 1,152 mutants using signature-tagged transposon mutagenesis on an infectious bacterial artificial chromosome… Show more

“…However, there was no evidence for the membrane fusion deficit that might have been predicted from previous herpesvirus gL descriptions. In this regard, the viability of the gL mutants contrasted completely with our (24) and others' (27,34) inability to propagate MHV-68 gH mutants. It seems unlikely that gH alone and gH/gL participate independently in fusion, as they appear to be conformationally quite different (Ͻ10% of gHspecific MAbs recognized both).…”

“…ORF64 is a tegument protein that is essential for MHV-68 replication (22). Because of technical difficulties in expressing fulllength ORF64, we analyzed fragments of the ORF64 coding sequence and identified two ZAP-responsive elements (Fig.…”

“…Expression of RTA is necessary and sufficient to trigger reactivation into lytic replication in latently infected cell lines (16)(17)(18)(19). RTA activates transcription of multiple genes expressed in the lytic phase (16,17,20,21), some of which are essential for lytic replication (22).…”

“…Open reading frame 24 (ORF24) is conserved among all beta-and gammaherpesviruses. Both ORF24 of MHV-68 and its human cytomegalovirus homolog (UL87) have previously been identified as being essential for lytic replication by genome-wide mutagenesis (7,19,25). ORF24 of MHV-68 is 39% and 26% identical to the Kaposi's sarcoma-associated herpesvirus and Epstein-Barr virus homologs, respectively (23).…”
