Identification of urinary metabolites of flavanone in the rat

Abstract: The urinary metabolites of flavanone in the rate were investigated using a gas chromatographic mass spectrometric method employing an OV-1 capillary column. Forty-three metabolites were detected and the most prominent of these were identified. The most common metabolic reactions were reduction of the keto group and hydroxylation at the 3- or 6-positions. Little hydroxylation occurred in ring B. The major metabolites were flavan-4 alpha-ol, trans-3-hydroxyflavan-4 beta-ol, 6-hydroxyflavanone and 6-hydroxyflavan… Show more

“…However, only a small amount of hydroxylation occurred in the B-ring. 12) 7-Hydroxyflavanone has been isolated from a number of sources including Spatholubus suberectus, Dalbergia cochinchinensis, and Virola surinamensis and is associated with biological activities including, antifungal, 13) 20S proteasome inhibitory activity, 14) and aromatase inhibitor properties. 15) Microorganisms are good predictors of mammalian drug metabolites with the ability to yield sufficient quantities of metabolites for structure analysis and further pharmacological evaluation.…”

“…8) Present study underlined the importance of using microbial models to mimic mammalian metabolism of xenobiotics. The completely characterized microbial metabolites of 1 could thus, be utilized to confirm tentatively identified mammalian metabolites 5) due to lack of material and also to carry out further pharmacological studies.…”

“…7) As many as 43 urinary metabolites of flavanone in the rat were detected by a gas chromatographic mass spectrometric technique and the most prominent compounds among them were identified. 8) The main transformations observed were hydroxylation at the 3-or 6-position and the reduction of the keto group. However, only a little hydroxylation occurred in B-ring.…”
