Identification of Three Subunits of the High Affinity ω-Conotoxin MVIIC-sensitive Ca2+ Channel

Abstract: N-, P-and Q-type voltage-dependent Ca 2؉ channels control neurotransmitter release in the nervous system and are blocked by -conotoxin MVIIC. In this study, both a high affinity and a low affinity binding site for -conotoxin MVIIC were detected in rabbit brain. The low affinity binding site is shown to be present on the N-type Ca 2؉ channel. Using optimized conditions for specific labeling of the high affinity -conotoxin MVIIC receptor and a panel of subunit specific antibodies, the molecular structure of the … Show more

“…Purification of cardiac Ca 2þ channels labeled by dihydropyridine Ca 2þ antagonists identified subunits of the sizes of the a1, a2d, b, and g subunits of skeletal muscle Ca 2þ channels (Chang and Hosey 1988;Schneider and Hofmann 1988;Kuniyasu et al 1992), whereas immunoprecipitation of Ca 2þ channels from neurons labeled by dihydropyridine Ca 2þ antagonists revealed a1, a2d, and b subunits but no g subunit ). Purification and immunoprecipitation of Ntype and P/Q-type Ca 2þ channels labeled by v-conotoxin GVIA and v-agatoxin IVA, respectively, from brain membrane preparations also revealed a1, a2d, and b subunits but not g subunits (McEnery et al 1991;Martin-Moutot et al 1995;Witcher et al 1995a;Liu et al 1996). More recent experiments have unexpectedly revealed a novel g subunit (stargazin), which is the target of the stargazer mutation in mice (Letts et al 1998), and a related series of seven g subunits is expressed in brain and other tissues (Klugbauer et al 2000).…”
