Identification of the Soybean Allergenic Protein,Gly mBd 30K, with the Soybean Seed 34-kDa Oil-body-associated Protein

Abstract: The soybean allergenic protein, Gly m Bd 30K [Ogawa et al., J. Nutr. Sci. Vitaminol., 37, 555-565 (1991)] which is most strongly and frequently recognized by the IgE antibodies in sera of soybean-sensitive patients with atopic dermatitis, has been characterized. The allergen was isolated from the crude 7S-globulin fraction as an oligomeric form with a molecular weight of more than 3000,000 by gel-filtration chromatography. On two-dimensional gel electrophoresis, the native oligomeric allergen had an isoelectri… Show more

“…The 7S complex is generally a trimer with MW 150-200 kD, whereas 11S is a hexamer with MW 300-400 kD [14]. In the 7S fraction, or b-conglycinin, several IgE-binding proteins have been identified and some of them have been purified and characterized: Gly m Bd 28K, Gly m Bd 30K, Gly m Bd 60K [15,16]. Sequence homology has been reported between these allergens and the thiol proteinase family [17], and with the 49 Detection and identification of a soy protein component that cross-reacts with caseins from cow's milk allergen Der p1.…”

“…Soy protein is a well-known allergenic protein, and the 34 kDa protein is a well identified allergenic protein in soy protein (Ogawa et al , 1993). Most patients who can not take soy protein are sensitive to 34 kDa protein.…”

“…In this study, we used the following primary antibodies against the major soybean allergens listed in Table 2: anti-7S globulin (rabbit polyclonal antibody) (12), anti-Gly m Bd 30K (mouse monoclonal antibody) (13), anti-Gly m Bd 28K (mouse monoclonal antibody) (14), anti-Gly m 4 and anti-Gly m 3 (rabbit polyclonal antibodies), and anti-soybean trypsin inhibitor (rabbit polyclonal antibody; Chemicon, Temecula, CA). All antibodies used in this study except for antisoybean trypsin inhibitor were prepared by us or other researchers.…”
