Identification of the porcine paramyxovirus LPMV matrix protein gene: comparative sequence analysis with other paramyxoviruses

Abstract: The complete nucleotide sequence of the gene encoding the matrix protein (M) of the porcine paramyxovirus LPMV has been determined. The gene is 1376 nucleotides long including 5' and 3' non-coding sequences with a protein-coding sequence of 1107 nucleotides. The deduced protein, containing 369 amino acids with a calculated Mr of 41657, is hydrophobic overall with a net positive charge of +17.5. Comparative sequence analysis revealed high amino acid homology to other paramyxovirus M proteins, with the highest d… Show more

“…This virus was originally isolated in Mexico from brain tissue of a pig exhibiting clinical signs typical of LPMV infection 10 and has been characterized at protein and genomic levels. 1,2,4,11,12 Following receipt of this isolate at the Veterinary Sciences Division, the virus was purified by 3 passages at limiting dilution in continuous pig kidney cell cultures (PK/ 15), and a working pool was prepared. The infectious titer of this working pool was calculated to be 10 7.50 TCID 50 /0.1 ml, following inoculation of serial 10-fold dilutions in PK/ 15 cell cultures.…”

“…9 Portions of the genome of LPMV have recently been sequenced, and some homology between LPMV and the mumps virus group of paramyxoviruses has been reported. 2,4,12 Although clinical signs, histopathology, and tissue distribution of virus following naturally occurring LPMV infection of pigs have been reported, 9 there are few detailed studies of the pathogenesis of the porcine paramyxovirus following experimental infection by natural routes. 9 Here, we report the results of a detailed sequential study of virus distribution in tissues of pigs experimentally infected with LPMV by a natural route, as determined by immunofluorescence staining and virus isolation techniques.…”

“…The generation and identification of LPMV genomic cDNA clones have been described earlier [3]. Selection of clones from the eDNA library were made by hybridization of [32p3 labelled random primed reverse transcribed viral RNA and Northern blot analysis (not shown).…”

“…Recently the structural proteins of LPMV were identified and in particular the phosphoprotein of LPMV migrated to a similar position of 52 kDa in SDS-PAGE as that in the case of the subgroup in the paramyxovirus genus consisting of mumps virus (MuV), simian virus 5 (SV5), parainfluenza 2 and 4 (PIV2 and 4), and Newcastle disease virus (NDV) [33]. The sequence of the matrix (M) protein gene of LPMV was determined and the closest relationship was found to be to MuV of 46% amino acid identity [3]. The family Paramyxoviridae is divided into three genera: Paramyxovirus, Morbillivirus, and Pneumovirus [17].…”
