Identification of the major chemical constituents and their metabolites in rat plasma and various organs after oral administration of effective Erxian Decoction (EXD) fraction by liquid chromatography‐mass spectrometry

Abstract: A simple and specific LC-DAD-ESI-MS/MS method has been developed and applied for the primary investigation of the chemical constituents absorbed or metabolized in vivo, after the rat oral administration of Erxian Decoction (EXD), a Chinese medicine prescription for menopausal syndromes. Through the online ESI-MS(n) analysis, a total of 35 compounds have been identified or tentatively characterized from the seven tested samples, and 13 of them were unambiguously identified through a direct comparison of the ret… Show more

“…Furthermore, liquid chromatography coupled with mass spectrometry (LC/MS n ) has been developed for qualitifying chemical constituents, including curculigoside, after oral administration of Erxian decoction to rats (Hu et al, 2010). However, these methods were not optimized for pharmacokinetic studies on curculigoside owing to their low specificity and sensitivity, as well as long run times.…”

“…To establish an integrated method for screening and analyzing the effective components in TCM, serum pharmacochemistry was proposed [1], which based on the hypothesis that active constituents were the components absorbed into blood. With the help of this method, the effective components of Erxian Decoction [2], She-Xiang-Bao-Xin pill [3], Dang-Gui-Bu-Xue decoction [4] were investigated by in vivo bioactive components fingerprint chromatogram.…”

“…According to the theory of plasma pharmacochemistry, only the constituents absorbed into blood have the chance to show effect and may be considered as potential bioactive components. By using this methodology, the effective components of many formulate had been observed such as Yin Chen Hao Tang , Shexiang Baoxin pill (Jiang et al, 2009), Er-Xian Decoction (Hu et al, 2010), Jitai tablets and Fufang Danshen tablets (Lv et al, 2010). In present study, the methodology of plasma pharmacochemistry was also applied to screen the bioactive components in HLJDT.…”
