Identification of the chlE gene encoding oxygen-independent Mg-protoporphyrin IX monomethyl ester cyclase in cyanobacteria

Abstract: The fifth ring (E-ring) of chlorophyll (Chl) a is produced by Mg-protoporphyrin IX monomethyl ester (MPE) cyclase. There are two evolutionarily unrelated MPE cyclases: oxygen-independent (BchE) and oxygen-dependent (ChlA/AcsF) MPE cyclases. Although ChlA is the sole MPE cyclase in Synechocystis PCC 6803, it is yet unclear whether BchE exists in cyanobacteria. A BLAST search suggests that only few cyanobacteria possess bchE. Here, we report that two bchE candidate genes from Cyanothece strains PCC 7425 and PCC … Show more

“…It once was believed that proteins encoded by these genes did not contribute to Chl biosynthesis (13); however, the recent finding that cyanobacterial bchE orthologs from two strains of Cyanothece restore BChl a biosynthesis in a bchE mutant of Rba. capsulatus demonstrates the activity of O 2 -independent BchE orthologs from oxygenic phototrophs (29).…”

“…There is also an O2-independent cyclase, BchE, which incorporates oxygen donated from water (196). Although BchE homologs (termed ChlE) have been found in some cyanobacteria (197), the O2independent cyclase is found mainly in anoxygenic phototrophic bacteria, and it will be described in the bacteriochlorophyll (BChl) section below. Pinta et al (198) assigned AcsF (aerobic cyclization system Fe-containing subunit) in the purple betaproteobacterium Rubrivivax gelatinosus to the O2-dependent reaction, and homologs were subsequently found in all oxygenic photosynthetic organisms investigated.…”

“…1). Firstly, there are two evolutionarily distinct types of Mg-protoporphyrin IX monomethyl ester cyclase (MgPME cyclase), one is a multi-subunit enzyme (Yamanashi et al, 2015;Chen et al, 2017) and the other is a single-subunit enzyme encoded by chlE (Yamanashi et al, 2015). To our knowledge, neither of the enzymes has been detected in complex algae (e.g., diatoms;Wilhelm et al 2006;Nymark et al 2009).…”
