Over recent years, technological advances have brought innovation in assisted reproduction to the agriculture. Fox species are of great economical interest in some countries, but their semen characteristics have not been studied enough. To advance the knowledge of function of fox spermatozoa, five samples were obtained by masturbation, in the breeding season. Kinetic analysis was performed using ISAS® v1 system. Usual kinematic parameters (VCL, VSL, VAP, LIN, STR, WOB, ALH and BCF) were considered. To establish the standardization for the analysis of samples, the minimum number of cells to analyse and the minimum number of fields to capture were defined. In the second step, the presence of subpopulations in blue fox semen was analysed. The minimum number of cells to test was 30, because kinematic parameters remained constant along the groups of analysis. Also, the effectiveness of ISAS® D4C20 counting chamber was studied, showing that the first five squares presented equivalent results, while in the squares six and seven, the kinematic parameters showed a reduction in all of them, but not in the concentration or motility percentage. Kinematic variables were grouped into two principal components (PC). A linear movement characterized PC1, while PC2 showed an oscillatory movement. Three subpopulations were found, varying in structure among different animals.

Se realizó el análisis de la motilidad con ayuda de un sistema computarizado de análisis seminal (CASA) en combinación con una tinción de fluorescencia Hoeschst 33259 para el estudio de la viabilidad y la morfología. Se realizó un análisis de medidas repetidas en el tiempo con el paquete estadístico SAS, empleando el método Proc Mixed. No hubo diferencias significativas (p > 0,05) en el porcentaje de motilidad total y motilidad progresiva para el primer y segundo eyaculado, pero sí hubo diferencias (p < 0,05) para la concentración, volumen y viabilidad; el primer eyaculado fue significativamente superior al segundo, independiente de la raza y la colecta. Las variables de cinemática espermática relacionadas con el movimiento progresivo y la velocidad (ALH, VAP, LIN) no difieren entre eyaculados, pero sí hay un efecto racial (p > 0,05) en el que la raza criolla es superior en estas variables, lo que podría sugerir que las células espermáticas de esta raza tienen la capacidad de recorrer mayor distancia en menor tiempo, comparado con los espermatozoides de las otras dos razas, en las condiciones del presente experimento. of total motility and progressive motility for the first and second ejaculates, but there were differences (p < 0.05) for concentration, volume, and viability; the first ejaculate was significantly superior to the second, independent of breed and collection. This study investigates the association of semen traits with boar fertility. The fertility outcome (farrowing rate - FR and total piglets born - TB) of 14 boars was obtained from a field trial conducted during 10 week of breeding period on a commercial farm using multiparous sows (n = 948) through single-sire mating with 2 × 10(9) motile sperm cells per artificial insemination (AI) dose. Sperm motion parameters, evaluated with computer-assisted semen analysis system in raw and stored semen at 17°C for 240 h, in addition to morphological sperm defects, measured on the collection day, were included in the analysis to determine which semen traits were important to discriminate the fertility potential of ejaculates from these boars. The data underwent multivariate cluster, canonical and discriminant analyses. Four clusters of boars were formed based on fertility outcome. One boar, with the lowest FR and TB values (89.7% and 11.98), and two boars, with the highest FR and TB values (97.8% and 14.16), were placed in different clusters. The other boars were separated in two distinct clusters (four and seven boars), including boars with intermediate TB (12.64 and 13.22) but divergent values for FR (95.9% vs 91.8%). Semen traits with higher discriminatory power included total motility, progressive motility, amplitude of lateral head displacement and cytoplasmatic droplets. Through multivariate discriminant analysis, more than 80% of the 140 ejaculates were correctly classified into their own group, showing that this analysis may be an efficient statistical tool to improve the discrimination of potential fertility of boars. Nevertheless, the validation of the relationship between fertility and semen traits using this statistical approach needs to be performed on a larger number of farms and with a greater number of boars.
