Identification of Sesquiterpene Synthases fromNostoc punctiformePCC 73102 andNostocsp. Strain PCC 7120

Abstract: Cyanobacteria are a rich source of natural products and are known to produce terpenoids. These bacteria are the major source of the musty-smelling terpenes geosmin and 2-methylisoborneol, which are found in many natural water supplies; however, no terpene synthases have been characterized from these organisms to date. Here, we describe the characterization of three sesquiterpene synthases identified in Nostoc sp. strain PCC 7120 (terpene synthase NS1) and Nostoc punctiforme PCC 73102 (terpene synthases NP1 and… Show more

“…Two such sesquiterpene synthases, Alr4685 and Npun_R3832, of Nostoc sp. PCC 7120 and Nostoc punctiforme PCC 73102, respectively, have previously been characterized in vitro (49). Consistent with these observations, heterologous Streptomyces expression of the corresponding sesquiterpene synthases gave rise to formation of sesquiterpene products.…”

“…S9). The most abundant corresponds to the newly discovered bacteriocin family (30,31), whereas the terpenes (32) are present in almost all of the 126 genomes. Even the genes encoding cyanobactins (33) were recovered from 10% of the dataset.…”

“…Germacradienol/geosmin synthase is atypical in that it consists of two terpene cyclase domains. Typical singledomain SCs characterized from Streptomyces and Nostoc include those involved in the synthesis of pentalenene, epi-isozizaene, avermitilol, germacrene A, 8a-epi-␣-selinene, (Ϫ)-␦-cadinene, and (ϩ)-T-muurolol (7)(8)(9)(10)(11)(12)(13). However, recent genome sequencing projects have revealed a large number of potential bacterial SC genes.…”
