Identification of potential mechanism and hub genes for neuropathic pain by expression‐based genome‐wide association study

Abstract: Neuropathic pain (NP) is a common pathological pain state with limited effective treatments. This study was designed to identify potential mechanisms and candidate genes using gene expression–based genome‐wide association study (eGWAS). All NP‐related microarray experiments were obtained from Gene Expression Omnibus and ArrayExpress. Significantly dysregulated genes were identified between experimental and untreated groups, and the number of microarray experiments in which each gene was dysregulated was calcul… Show more

“…Quantitative real‐time polymerase chain reaction (qRT-PCR) was conducted as reported previously [15]. In short, total RNA was extracted from snap-frozen DRG and spinal cord tissues using TRIzol reagent (Thermo Fisher Scientific, Waltham, MA, U.S.A.).…”

“…Western blotting was performed following the standard procedures as previously described [15] by using the following antibodies: C–C motif chemokine ligand 2 (CCL2) (rabbit, 1:1000, Novus, Littleton, CO); allograft inflammatory factor 1 (AIF1; rabbit, 1:1000, Novus); tissue inhibitor of metalloproteinase 1 (TIMP1) (rabbit, 1:1000, Sigma–Aldrich, St. Louis, MO) and β-actin (1:3000; Sigma–Aldrich). All Western blots were repeated at least three times.…”

“…GO and KEGG enrichment analyses uncovered that these functions were associated with the occurrence and development of NP. Additionally, complement and coagulation cascade [ 30 ], neuroactive ligand-receptor [ 31 ], and ECM-receptor interactions [ 32 ] were signaling pathways related to various functions mediated by NP. The genes associated with these pathways were also observed in our analysis.…”
