Identification of PLK1 as a New Therapeutic Target in Mucinous Ovarian Carcinoma

Abstract: Mucinous epithelial ovarian cancer (mEOC) is a rare subset of epithelial ovarian cancer. When diagnosed at a late stage, its prognosis is very poor, as it is quite chemo-resistant. To find new therapeutic options for mEOC, we performed high-throughput screening using a siRNA library directed against human protein kinases in a mEOC cell line, and polo-like kinase1 (PLK1) was identified as the kinase whose downregulation interfered with cell proliferation. Both PLK1 siRNA and two specific PLK1 inhibitors (onvans… Show more

“…PLK1 activates the SAC signal when inhibited [ 216 ]. PLK1 expression has been connected with disease prognosis and sensitivity to chemotherapy in ovarian cancer, which presents a potential biomarker or therapeutic target in combating resistance [ 201 , 216 , 219 , 220 ]. Studies have shown that inhibiting PLK1 in the presence of PTX [ 201 , 216 ] resensitized ovarian cancer cells to PTX and reduced cell proliferation, as well as inducing apoptosis in HGSOC cells with CCNE1 amplification.…”

“…CDK1 expression is elevated in a variety of cancers, often leading to unrestricted proliferation of malignant tumor cells [19][20][21]. PLK1 is a serine threonine kinase that plays a vital role in centrosome maturation, mitotic chromosome separation, and mitosis, and is closely related to the occurrence and development of malignant tumors [22][23][24]. As a kinase, CHEK1 also plays an important role in tumor progression by regulating the cell cycle [25,26].…”

“…Further large prospective studies are needed to confirm our data. OC represents a group of heterogeneous diseases [35][36][37][38][39][40]; therefore, further investigations are needed to evaluate the applicability of e-nose in detecting various OC types. The new perspective is the implementation of this device in the new collection of exhaled breath samples on a larger population to validate these results and build more robust classification models.…”
