Identification of Novel Bovine Cumulus Cell Molecular Markers Predictive of Oocyte Competence: Functional and Diagnostic Implications1

Abstract: The present study was undertaken to discover molecular markers in bovine cumulus cells predictive of oocyte competence and to elucidate their functional significance. Differences in RNA transcript abundance in cumulus cells harvested from oocytes of adult versus prepubertal animals (a model of poor oocyte quality) were identified by microarray analysis. Four genes of interest encoding for the lysosomal cysteine proteinases cathepsins B, S, K, and Z and displaying greater transcript abundance in cumulus cells s… Show more

“…Although a potential effect of the enzymatic method in cumulus cell gene expression profile cannot be ruled out, we thought that mechanical isolation of cumulus cells before ICSI could jeopardise the success of our patients' fertility treatment rendering our strategy unethical. Nevertheless, hyaluronidase has been used by other researchers in cumulus cell gene expression studies even in non-human mammals (cows) [2,3].…”

“…Several cathepsin transcripts, including CTSB and CTSZ, are inversely related to oocyte quality. In one study, cumulus derived from COCs of prepubertal heifers, a model of poor oocyte competency, contained more CTSB mRNA than COCs from mature cows (Bettegowda et al 2008). Oocyte quality and postfertilization development could be improved by exposing COCs to a membrane-permeable cathepsin inhibitor (Bettegowda et al 2008).…”

“…The quality of oocytes and the fate of embryos are determined by the maternal legacy (Bettegowda et al, 2008;Lechniak et al, 2008). It has been considered that some of the specific molecules, which are not well known, determine oocyte quality.…”

“…With that, FST could have a potential and functional role in embryonic development that affects embryo quality (Patel et al, 2007;Bettegowda et al, 2008). Therefore, it was hypothesized that FST supplementation would enhance the embryonic development when it was treated in culture medium during in vitro culture of porcine embryos after IVF.…”
