Identification of mutations in porcine STAT5A that contributes to the transcription of CISH

Abstract: Identification of causative genes or genetic variants associated with phenotype traits benefits the genetic improvement of animals. CISH plays a role in immunity and growth, however, the upstream transcriptional factors of porcine CISH and the genetic variations in these factors remain unclear. In this study, we firstly identified the minimal core promoter of porcine CISH and confirmed the existence of STATx binding sites. Overexpression and RT-qPCR demonstrated STAT5A increased CISH transcriptional activity (… Show more

“…Among these, STAT5A has been linked to cell specification, proliferation, differentiation, and survival at the molecular level [42]. Additionally, STAT5A has been found to play a role in immunity and growth by positively regulating porcine CISH gene transcription, and the SNP (g.566C > T) of STAT5A was associated with the piglet growth trait [43]. TGFB2 is a critical ligand in the TGF-β signaling pathway [44].…”

“…Animal growth parameters are important in livestock breeding programs. Some hormones that are associated with animal growth have been reported, including growth hormone receptor (GHR) [ 1 ], growth hormone-releasing hormone (GHRH) [ 2 ], pituitary-specific transcription factor-1 (PIT-1) [ 3 ], and signal transducer and activator of transcription 5A (STAT5A) [ 4 ]. The GHR acts as a transmembrane protein, and its primary function is to bind GH with great specificity and affinity.…”
